What is FOXO4-DRI?

A cell-penetrating peptide (14 amino acids) designed to disrupt the interaction between FOXO4 and p53 proteins, thereby preventing cellular senescence and enabling senescent cell clearance. Targets aging at the cellular level via senolytic mechanisms.

FOXO4-DRI is a cell-penetrating peptide that inhibits FOXO4-p53 interaction. This prevents p53-mediated activation of senescence pathways, allowing immune clearance of senescent cells.

Background & history

Discovered in 2017 by Dutch researchers at Erasmus University; showed remarkable anti-aging effects in mice. No commercial development in humans; remains experimental.

What the research says

Preclinical mouse studies show FOXO4-DRI reduces senescent cell burden and improves multiple aging-related outcomes, including kidney function and muscle mass. No human clinical trials have been completed. Mechanism is sound in animal models, but translation to humans is unproven. Safety profile in humans is entirely unknown.
